Focus on the Sample Surface

  1. Click the Navigate icon in the Workflow Toolbar.
  1. Click the Sample (default) button in the Focus Sample panel.
 
  1. Ensure that the Auto-compensate for fluid check box is selected.
  2. CAUTION: If the Auto-compensate for fluid box is checked and focus is performed in air, the tip will crash on engage.
Focus on the sample surface either with the Focus: Z Motor arrows or by rolling the trackball up or down while pressing the bottom-left button. This adjustment raises or lowers the vertical engage stage on which the SPM and optics are mounted.

 
  1. To move long distances hold both left trackball buttons down simultaneously and roll the trackball with high speed to lock the peak speed of motion. Release these two buttons to stop the motors. Be careful when performing this in the downward direction. Focus on the sample surface using the trackball with the bottom-left button depressed.
CAUTION: Because the command Focus Surface moves the scanner vertically, be careful when making this adjustment to ensure that the tip does not hit the sample surface.
 
  1. Add fluid.
 
  1. If necessary, Readjust the Laser Alignment.
 
  1. If necessary, Adjust the Photodetector.
NOTE: Fluid changes the optical path length as the angle of refraction changes, therefore, you must offset the focus position in Focus Surface by 1.3 times to a point below the surface when focusing in fluid. For example, if the surface is in focus with the Z motor positioned at –1000 µm, you must move the Z motor position down until it is at –1300 µm.
